The process of a sibling DNA test starts with the collection of buccal (mouth) swabs from each of the potential siblings. These swabs gather cells that hold unique genetic information in the form of DNA, similar to a fingerprint. In our lab, we carefully extract the DNA from the cells and amplify specific DNA regions using a technique called PCR (polymerase chain reaction) for a comprehensive examination. We then compare the DNA patterns of the siblings. A statistical analysis is performed based on the match type typically seen in biologically related siblings, generating a “full sibling index” and “half sibling index.” The higher the index value over 1.0, the greater the likelihood of a biological sibling relationship. Conversely, a lower index value below 1.0 suggests a lower probability of a biological sibling relationship.
