Ever wondered if there’s a genetic reason behind relationship dynamics? Research points to a gene called AVPR1A, sometimes called the “male pair-bonding” gene, which produces a protein known as arginine vasopressin receptor 1A.
One particular variation in the AVPR1A gene, the RS3 334 allele, has sparked interest due to its possible role in shaping how men bond in relationships.
Men who carry the RS3 334 allele:
- Might find it challenging to form deep bonds with partners.
- Have a higher likelihood of marital problems or divorce.
- Often prefer cohabitation over marriage compared to those without this allele
Interestingly, partners of these men often report feeling less warmth and agreement in their relationships.
The Genetics
Accurately identify if you carry the RS3 334 variant of the AVPR1A gene, a variant known to influence pair-bonding behaviours in males. Each person inherits two variants of this gene – one from each parent.
Research indicates that about 40% of Caucasian men have at least one copy of the AVPR1A RS3 334 allele. Men with just one RS3 334 allele are less likely to form deep bonds with their partners. Those with two copies are at a heightened risk—34% chance—of facing marital challenges or potential divorce, compared to the 15% risk in men with different AVPR1A gene variations. Additionally, these men are more likely to prefer cohabitation over marriage.
While the RS3 334 allele is linked to weaker bonding in men, the RS3 334 allele doesn’t seem to impact bonding in the same manner for women. Instead, five other changes in the AVPR1A gene are associated with an increased likelihood for infidelity in women.

Vasopressin is a hormone that acts as our body’s “chemical messenger.” When released into the bloodstream, it helps our kidneys retain water and helps our bodies regulate blood pressure.
Vasopressin can also travel directly to the brain, where it can influence our social interactions and intimate behaviours, including tendencies like infidelity.
Arginine vasopressin receptor 1A comes into play by acting as the bridge between our bodies and vasopressin. It’s the receptor’s job to “catch” vasopressin. Once bound, the receptor amplifies and transmits the hormone’s instructions inside our cells, guiding our body’s varied responses.
